How will Australia assess sector applicants for their skilled visa?

How will Australia assess experience and specialization under the new priority criteria for farming, fishing, and mining, compared with roles in construction, healthcare, teaching, defence, and law enforcement? Will experience in a specific position be as important as years of expertise?

The quick answer is no, experience in a specific position is not the deciding criteria.
Priority is determined by the nominated occupation's ANZSCO code in the application, regardless of the applicant's specific experience. The ministerial direction sets a range of ANZSCO codes that list the recognised occupations for each relevant sector. Outside of the ANZSCO code (which in some cases defines a group of occupations, and in some cases a specific occupation) there is no consideration for specialisation.
