After some research on the Expression of Interest (EOI) system, I feel a little confused about the effectiveness of the EOI on screening the most qualified candidates of PNPs. What are some standards that the EOI uses to give grades? How can candidates get a higher score in a fact-based question system with no human involvement in the grading procedure?

Different provinces have different scoring system for their EOI. If you look at Manitoba, for example, the pertinence of the project (and how the applicant can be full time involved in it) is more important than the individual score. I suggest contacting an immigration lawyer to be best guided. Do keep in mind that a lot of PNPs are on soft launch and their process is a moving target as we speak, so if you wait for information to appear on
forums, it could already be outdated.
